Araştırmalarımda genel ilgi alanım: embriyonik ve indüklenmiş kök hücre sistemi kullanarak kalp-damar gelişimi ve hastalıklarının moleküler temellerinin araştırması ve tedavi yöntemleri geliştirilmesidir. Özel alt başlıklar ise: • Embriyonik kök hücrelerden kalp kök hücresi ve kardiyomiyosit elde edilmesi • Kalp kök hücrelerinin farklılaşma ve kendini yenilemesinde gerekli sinyal ileti mekanizmalarının tanımlanması • Kalp ve damar hastalıkları tedavisinde kullanılmak üzere yeni kalp doku mühendisliği uygulamalarının geliştirilmesi • Hastaların deri dokusundan uyarılmış pluripotent kök hücre elde edilmesi ve in vitro modelleme yapılarak hastalık mekanizmasının araştırılmasıdır.

Doğal bileşiklerin aktif maddelerinin izolasyon, sentezleme metodları ve teknikleri. • Katalitik asimetrik indüksiyon reaksiyonlarının mekanizmalarının incelenmesi ve bunlardan çıkarak biyoljik sistemlerde aktif olabilecek ilaçlarin geliştirilip sentezleme yöntemleri.

In cancer therapy, toxicity in healthy tissue is a common side effect of current treatment modalities and tumor recurrence is almost inevitable. There is therefore an urgent need to develop novel therapies that act in a robust and cancer-selective manner. In line with this, main focus of our research is developing novel targeting therapy approaches for cancer treatment. One of the succesfull delivery strategy for targeted therapeutics without the limitations of current chemotherapeutic agents, is using stem cells as therapeutic cargo carriers since they have known with their migration capacity towards the tumor sites even including microdeposits. Furthermore, stem cells can be collected individually and engineered to secrete therapeutics which can enable their clinical translation from bench side to bed side. Recently, we are working on engineering stem cells to secrete anti-cancer therapeutics including targeted toxins for the treatment of high grade gliomas and other malignancies. For this aim, we combine bioimaging markers (fluorescence proteins and firefly luciferases) and therapeutic molecules in stem cells using lentiviral vectors. Following pre- and post-implantation of these therapeutically engineered stem cell cargos in mice bearing bioimagable tumors, we then analyse tumor regression by fluorescence/bioluminescence imaging tools and subsequent tissue processing. Furthermore, we investigate the molecular mechanism leading to cancer cell death in culture and to loss in tumor mass in treated mice upon therapeutic stem cell implantation.
